Counting Methodology and Sensitivity
SA is the most sensitive mail-in kit on the market. As the original mail-in test kit, we’ve been perfecting this process for over 30 years.
Semen Analysis
A standard semen analysis is performed to evaluate fertility and/or health of the male organs, including:
Semen volume
Total volume of semen specimen received.
Sperm Count / Concentration
Measures the total number of sperm. It is recorded in millions of sperm per milliliter of semen. A normal count is greater than (≥) 20 million per ml. Lower than normal is still viable in many fertility procedures.
Motility
The percentage of sperm moving and how well they move. Normal semen will have at least 50% motile sperm.
Morphology
The form and structure of a single sperm. Sperm are evaluated by both WHO (World Health Organization) 1994 criteria, and Kruger (Fertil Steril 49:112, 1988) strict criteria. Normal Kruger is ≥14, Normal WHO is ≥60.

How Sperm Counting Has Evolved
Historically, semen analysis (SA) involved an in-office process where patients provided a sample for direct examination under a standard light microscope.
The Hemacytometer Advantage
In clinical laboratories, including ours, hemacytometers are the gold standard for accurate sperm counting. These tools feature a mirrored grid with a precise volume of 0.9 microliters per side. When fully examined, a single sperm observed in this volume corresponds to:
1 sperm = 1,111 sperm/mL
This high sensitivity makes hemacytometry the most refined technique available for laboratory sperm quantification.
Sperm Washing for Cryopreservation
Sperm are separated from seminal fluids using a centrifuge. This is a standard procedure used for many infertility treatments.
